Spectrophotometry 

In order to measure the enzyme activity of Lac, we use ABTS as the substrate.
The assay mixer contained 1mM ABTS and 0.1M sodium citrate buffer, pH3.0. 
Oxidation of ABTS WAS followed by absorbance increase at 420nm.
